Sulforaphane is the most potent natural activator of the Nrf2 pathway — the master regulator of cellular antioxidant defenses. Avmacol provides broccoli sprout extract with myrosinase enzyme to ensure glucoraphanin converts to active sulforaphane in the gut. Decades of research link sulforaphane to cancer chemoprevention, detoxification enhancement, and anti-inflammatory effects.
Avmacol Sulforaphane scores 76 — the second-highest in the longevity category — because sulforaphane has one of the strongest and broadest evidence bases of any natural compound relevant to aging. The Nrf2 pathway is central to cellular defense, and sulforaphane is its most potent natural activator. Dozens of human trials (not just one or two) support its bioactivity. Avmacol includes myrosinase enzyme, which is critical for converting glucoraphanin to active sulforaphane — many competing products lack this. No hype penalty because the research is extensive and from independent academic groups.
Sulforaphane is the most extensively studied isothiocyanate, derived from glucoraphanin in cruciferous vegetables (especially broccoli sprouts). Egner et al. (2014, Cancer Prevention Research) showed sulforaphane-rich broccoli sprout beverage enhanced detoxification of airborne pollutants by 61% in a randomized trial in China (n=291). Singh et al. (2014, Cancer Prevention Research) demonstrated sulforaphane modulated Phase II detoxification enzymes in a dose-dependent manner. Fahey et al. (2017, Nutrients) reviewed the extensive evidence for sulforaphane as a Nrf2 activator — the master switch for >200 cytoprotective genes. Houghton et al. (2013, PLOS ONE) showed sulforaphane had anti-inflammatory effects comparable to synthetic Nrf2 activators. The longevity connection comes through Nrf2's role in oxidative stress defense, DNA repair, and mitochondrial function — all central to the aging process. Unlike many longevity compounds, sulforaphane has substantial human evidence across dozens of trials.

Sulforaphane is not traditionally marketed as a 'longevity' supplement, and the anti-aging framing is newer than the established cancer chemoprevention research. The amount of active sulforaphane generated from broccoli sprout extract varies between individuals based on gut microbiome composition. Fresh broccoli sprouts provide the same compound and are cheaper. GI side effects (gas, bloating) are common at higher doses. The Nrf2 activation story is compelling for cellular defense but direct lifespan extension in humans has not been studied.
